ASM cells were serum starved for 24 hours and then treated with medium alone, 10 ng/ml IL-1β, 10 ng/ml TNFα, 10 ng/ml IFNγ, 50 ng/ml IL-13, 1 μM dexamethasone or 1 μM salmeterol for 24 hours. Cells were loaded with [3H]-myo-inositol and IPx production following 100 μM Histamine, 0.1 μM bradykinin or 10 mM Sodium Fluoride stimulation was determined as described [30]. The IPx production in the medium alone treated cells was normalised to 100% and the other treatment groups determined relative to this. Data represents the mean response +/- S.E.M, n = 9 (three independent experiments for 3 donors). Data within groups was compared by ANOVA and post hoc Dunnett's Multiple Copmarison Test (+p < 0.05) or T-Test (*p < 0.05, **p < 0.01). | |||
