Resolution: standard / high Figure 6.
In vitro stimulation of human fibroblasts. A. Recombinant TNF-α increases the mRNA expression of tenascin-C in foetal lung
fibroblasts (MRC-9). Expression peaks at 9 hours after stimulation. The results are
from one experiment. B. Recombinant TNF-α enhances the mRNA expression of tenascin-C
also in human primary fibroblasts. TNF-α along with IFN-γ has a synergic effect, which
further enhances the expression of tenascin-C mRNA. Control mRNA expressions of tenascin-C
are adjusted to 1 and other expressions are shown as the fold change compared to the
control. C. An immunoblot showing enhanced expression of protein for tenascin-C (Mr 290,000 band) in the primary fibroblasts (the first two lanes) and in the cell culture
supernatant (the latter two lanes) after stimulation with TNF-α and IFN-γ for 18 h.
Below SYPRO Ruby staining of the immunoblot confirming equal loading and transfer
of proteins from cell lysate samples.
